Utilization of oxygen and reduced nicotinamide adenine dinucleotide phosphate by human placental microsomes during aromatization of androstenedione.

چکیده
The addition of androstenedione and NADPH to human placental microsomes results in a marked increase in the rate of 0 2 and NADPH utilization concomitant with estrogen formation. Initially, 5 to 12 moles of O2 and an equivalent amount of NADPH are consumed per mole of estrogen formed, the ratio varying from one microsomal preparation to another. In the presence, however, of 10 to 20 mM KCN, 3 moles of 02, and 3 moles of NADPH are consumed per mole of estrogen formed. This ratio was invariant even when a concentration of KCN sufficiently high to inhibit aromatization was employed. Both NADPH and O2 are required for the conversion of 19-hydroxyandrostenedione to 19-oxoandrostenedione, suggesting that a second hydroxylation is responsible for this conversion rather than oxidoreduction of the 19-alcohol. These results are consistent with previous data suggesting three successive hydroxylations catalyzed by a mixed function oxidase(s) are required during Cl9 steroid aromatization. The rate of appearance of estrogen intermediates was examined in an effort to determine to what extent the accumulation of these compounds might account for the high rate of O2 consumption observed in the absence of KCN. It was determined that essentially all of the 02 consumed could be accounted for by accumulation of 19-hydroxyandrostenedione and 19-oxoandrostenedione and estrogens. In the presence of KCN, the rates of formation of 19-hydroxyandrostenedione and 19-oxoandrostenedione were greatly inhibited under steady state conditions while the rate of aromatization was only slightly decreased. متن کاملEnhancement of DMA Chain Breakage by Bleomycin A2 in the Presence of Microsomes and Reduced Nicotinamide Adenine Dinucleotide Phosphate1
Chain breakage in DMA induced by bleomycin A (BLM) was enhanced more than 150-fold by the reduced nicotinamide adenine dinucleotide phosphate-dependent elec tron transport system of rat liver microsomes. However, the enhancement effect on DMA was partially reduced by the preincubation of BLM with the microsomal systems. BLM-Cu2' was found to stimulate considerably micro somal reduced nicotinami...
متن کاملInitial biotransformations of daunorubicin to aglycones by rat liver microsomes.
Anaerobic incubations of rat liver microsomes convert daunorubicin into at least six or seven aglycones in the presence of reduced nicotinamide adenine dinucleotide phosphate or a reduced nicotinamide adenine dinucleotide phosphate-generating system. The partial identification of some of the aglycones is based on cochromatographs of chemically synthesized derivatives by isocratic high-pressure ...
متن کاملVerifying of Participation of Nitric Oxide in Morphine Place Conditioning in the Rat Medial Septum Using Nicotinamide Adenine Dinucleotide Phosphate-Diaphorase (NADPH-d)
Background: Role of nitric oxide (NO) in morphine-induced conditioned place preference (CPP) has already been proposed in the rat medial septum (MS), but no molecular evidence has been provided to clear this fact. Methods: Effects of intraseptal injections of L-arginine and/or NG-nitro-L-arginine methyl ester (L-NAME) on morphine place conditioning in Wistar rats were examined. Morphine (2.5-7....

SUMMARY Different functional and structural properties of rat liver microsomes were studied during hepatocarcinogenesis induced by 0.25% m-ethionine. During the first to fourth months of ethionine feeding, great decreases of cytochrome P-450 content, reduced nicotinamide adenine dinucleotide phosphate-dependent lipid peroxidation, and aminopyrine demethylase activity occurred. No changes in the...
